You could say, for instance, Was interessiert Sie am meisten? ---> = What are you interested in most? or What are you most interested in You can't say "Was interessieren Sie". It doesn't make any sense. However, you could say "Für was interessieren Sie sich?" and then it would mean "What are you interested in".
